The lucrative tournament is set to take place in October, but Prior will not travel with Sussex after they came to an agreement with the England and Wales Cricket Board.
Prior, 27, has a busy winter schedule to fulfill as England travel to South Africa for the ICC Champions Trophy in late September before a one-day and Test series against South Africa between November and January.
Despite initial concerns over his work behind the stumps, the wicketkeeper has become an integral part of the England team after an impressive Ashes series with the bat and the gloves.
Mark Robinson, professional cricket manager for Sussex, was respectful of Prior's decision to focus on international cricket.
'He [Prior] obviously is desperate to help his county do well in what is a major competition and he's also aware of the other potential avenues that could be opened up by him doing well as an individual in India, such as the Indian Premier League,' he said.
'When talking with him, the one thing that kept coming out was his desire to be as fit and as fresh as possible to represent his country. As this was his only rest period, it was the right thing to do and we all at Sussex must respect Matt's dream to continue his England ambitions.'
England's cricket managing director, Hugh Morris, said he was appreciative of Sussex allowing Prior to miss the Champions League: 'I'd like to thank Sussex for their continued support of the England team this season. We appreciate their understanding in making this decision.'
One Sussex and England player who will be travelling to India is Luke Wright, with the all-rounder included in the Shark's 15-man squad.
Sussex won the domestic Twenty20 Cup and will be joined on the sub-continent by runners-up Somerset.
The tournament gets underway on October 8th with teams from Australia, South Africa, New Zealand, India, Sri Lanka, and the West Indies also taking part.
