A spokesperson for Scotland Yard confirmed that officers were called to the Piccadilly branch of Waterstone's after the incident but said Lewis did not require hospital treatment.
The singer's own spokesman said she had been signing autographs at the bookstore for around 90 minutes when 'a guy came up and punched her to the side of the head'.
The Bleeding Love star was 'understandably badly shaken', he added.
Waterstones have since issued a statement regarding the matter, saying: 'There was an incident at our Leona Lewis signing this afternoon and the event was immediately stopped.'
'We are now doing everything we can to support the police in their enquiries.
'We take security at our book signings extremely seriously and this is the first time anything of this nature has happened.'
The statement added: 'We would like to thank Leona for her professionalism and we are extremely sad that this event was ruined for her, and her fans, many of whom queued from the early hours of the morning to meet her.'
Lewis, 24, was signing copies of her autobiography Dreams, due for release next month.
The winner of The X Factor in 2006, she is a three-time Grammy nominee and has sold more than five million copies of her debut album Spirit.
